How it works
1. Prepare the sample according to the simple guidelines (see manual), e.g. dilute blood or saliva, or add swab material directly.
2. Combine Genotyping PCR Mastermix + Primers & Probes Mix + sample (total volume 20 µL).
3. Run the real-time PCR program (95 °C / 60 °C, 50 cycles) followed by a melting-curve analysis (45–85 °C) in the FAM channel.
Analysis:
4. Interpret the melting peaks:
Homozygous wild-type → single peak
Heterozygous → double peak
Homozygous mutant → single peak at lower temperature
Minor shifts in melting temperature (±0.5–2 °C) can occur depending on sample composition (salt, pH) – this is normal and does not affect genotype calling.
Example data: blue curve → heterozygous genotype; green curve → homozygous wild-type genotype
